Dietary fat has dramatic effects on liver metabolism. These effects can be beneficial, but they can also contribute to the onset and progression of chronic disease, e.g., coronary artery disease and diabetes. Many effects of dietary fatty acids can be ascribed to changes in gene expression through regulating the activity or abundance of key transcription factors, like the peroxisome proliferator activated receptor-alpha (PPAR-alpha) and the helix-loop helix transcription factor, sterol regulatory element binding protein-1c (SREBP-1c). These transcription factors play a major role in hepatic as well as whole body carbohydrate, lipid and cholesterol homeostasis. As such, our research has focused on defining the molecular basis for fatty acid regulation of these transcription factors as well as their contribution to chronic disease.
